Title:
Extracellular ATP as a transmitter; physiological role, regulation and molecular pharmacology of P2X1 receptors.

Abstract:
ATP is released from neurons, glia, in response to shear stress in blood vessels, and from damaged cells and can act at P2X receptors. P2X receptors comprise a distinct family of ligand gated ion channel with two transmembrane domains, intracellular amino and carboxy termini and a large extracellular loop. The seminar will focus on (i) the use of knockout mice to determine the physiological roles of P2X1 receptors in control of arterial tone and blood clotting, (ii) show how fluorescent recovery after photobleaching (FRAP) of GFP tagged P2X1 receptors can be used to study trafficking of the receptors and (iii) the use of site directed mutagenesis to develop an understanding of the molecular basis of ATP binding at the P2X1 receptor.
